耐低溫石油烴降解菌的篩選及降解性能研究

Screening and Degradation Performance of Cold-Adapted Petroleum Hydrocarbon Degrading Bacteria

  • 摘要: 針對低溫地區石油汙染土壤的生物修復,從遼河油田興隆臺土樣中篩選出的耐低溫石油烴降解菌群X-3✊,經16S rDNA序列分析鑒定,其中包括2株假單胞菌(Pseudomonas sp.)、1株節桿菌(Arthrobacter sp.)、1株紅球菌(Rhodococcus sp.)🍄‍🟫🌳、2株芽孢桿菌(Bacillus sp.)和1株蒼白桿菌(Ochrobactrum sp.)🏊‍♂️🧖🏽‍♂️。通過實驗優化X-3的培養條件👨‍👨‍👦,結果表明:在溫度10~15 ℃😑、接種量3%(質量百分比)🧓🏿、石油烴最適濃度10 g·L–1、最適氮磷比10∶1、最適pH為7的條件下🚗,X-3對石油烴的降解率從最初的47.75%增加到了60.07%。氣質聯用儀分析降解前後石油烴組分表明,X-3對C10~C21的石油烴均有一定程度的降解,並且隨著碳原子數的增加🤺,降解菌對石油烴的利用率逐漸降低,其中C16—4CH3的降解率僅為25%,表明含支鏈的烴類汙染物的生物降解比直鏈烷烴類更困難♛。

     

    Abstract: In view of the bioremediation oil-contaminated soil in low-temperature areas, the low-temperature resistant petroleum hydrocarbon degrading bacteria group X-3 was selected from Xinglongtai soil sample of Liaohe Oilfield and was identified by 16S rDNA sequence analysis. It was found that X-3 included 2 Pseudomonas sp., 1 Arthrobacter sp., 1 Rhodococcus sp., 2 Bacillus sp., and 1 Ochrobactrum sp. The culture conditions of X-3 were optimized through experiments. The results showed that under the conditions of temperature 10-15 ℃, the inoculation amount was 3% (mass percent), the optimum concentration of petroleum hydrocarbon was 10 g·L–1, and the optimum nitrogen phosphorus ratio was 10∶1. Under the condition of the optimum pH value of 7, the degradation rate of petroleum hydrocarbon by X-3 increased from 47.75% to 60.07%. GC-MS analysis of petroleum hydrocarbon components before and after degradation showed that X-3 could biodegrade the n-alkanes ranging from C10to C20 to a certain extent and the utilization rate of petroleum hydrocarbon by degrading bacteria decreased gradually with the increase of carbon atom number, in which the degradation rate of C16—4CH3 was only 25%, indicating that the biodegradation of branched hydrocarbon pollutants was more difficult than that of straight chain alkanes.
